CONy 2023 | Is α-synuclein toxicity the core element of PD?

Tiago Outeiro, PhD, University Medical Center Göttingen, Göttingen, Germany, discusses the role of α-synuclein in the pathogenesis of Parkinson’s disease (PD). The most pathological hallmark of PD is Lewy bodies (LBs), which are intraneuronal inclusions that contain immunoreactive α-synuclein aggregates. Some studies suggest that LBs are the cell’s protective mechanism to prevent intracellular protein aggregate accumulation, while other studies indicate LBs have a pathogenic role in PD. Therefore, whether α-synuclein toxicity is the core element of PD remains an area of controversy. This interview took place at the 17th World Congress on Controversies in Neurology (CONy) in Dubrovnik, Croatia.
