AES 2022 | Maladaptive myelination: a new therapeutic target in epilepsy and beyond?

In a recent study led by Juliet Knowles, MD, PhD, Stanford University, Palo Alto, CA, it was demonstrated that absence seizures cause maladaptive myelination in the brain that in turn promotes epilepsy progression in animal models of generalized epilepsy. This discovery could provide an exciting new therapeutic target for the treatment of epilepsy, however more work is needed to evaluate how to prevent maladaptive myelination whilst preserving healthy myelin plasticity. Although it has only been demonstrated within the context of generalized epilepsy, Dr Knowles suggests that this process of maladaptive activity-dependent neural network adaptation could occur in other types of epilepsy and even other types of neurological disorders. This interview took place at the American Epilepsy Society (AES) Annual Meeting 2022 in Nashville, TN.
